Amitriptyline — Tricyclic antidepressant (TCA). Study the mechanism, listed uses and warnings from the US label on one sourced card.
Generic name
Amitriptyline
Class
Tricyclic antidepressant (TCA)
Mechanism
Inhibits norepinephrine and serotonin uptake in adrenergic and serotonergic neurons. Its mechanism of antidepressant action in humans is not known; reuptake inhibition is a proposed explanation.
Antidepressants increased suicidal thinking and behavior in children, adolescents and young adults.
Warnings
Concomitant MAOIs (contraindicated); Cisapride (QT prolongation risk); Acute recovery phase after MI
Interactions
MAOIs; Topiramate; CYP2D6 inhibitors

Mechanism: Mechanism of mood elevation unknown; appears to interfere with transport, release and storage of catecholamines.
On both cards: Common side effects — Dry mouth; Blurred vision; Constipation; Urinary retention · Interactions — CYP2D6 inhibitors
Only on the Nortriptyline card: Used for — Relief of depressive symptoms in adults · Common side effects — Hypotension · Warnings — Psychiatric MAOIs are contraindicated with nortriptyline and within 14 days of switching in either direction; Do not start nortriptyline in a patient receiving linezolid or intravenous methylene blue because of serotonin-syndrome risk · Interactions — Cimetidine; Anticholinergic / sympathomimetic drugs
Mechanism: Exact mechanism unknown; believed to potentiate serotonergic and noradrenergic activity in the CNS.
On both cards: Common side effects — Dry mouth; Constipation · Interactions — MAOIs
Only on the Duloxetine card: Used for — Major depressive disorder in adults; Generalized anxiety disorder in adults and children aged 7 years and older; Diabetic peripheral neuropathic pain in adults; Fibromyalgia in adults and adolescents aged 13 years and older; Chronic musculoskeletal pain in adults · Common side effects — Nausea; Somnolence; Hyperhidrosis · Warnings — Hepatotoxicity; Orthostatic hypotension, falls, syncope; Serotonin syndrome · Interactions — Potent CYP1A2 inhibitors; Potent CYP2D6 inhibitors
Mechanism: Relatively weak inhibitor of neuronal norepinephrine and dopamine reuptake; does not inhibit serotonin reuptake or monoamine oxidase.
On both cards: Common side effects — Dry mouth · Interactions — MAOIs
Only on the Bupropion card: Used for — Major depressive disorder in adults · Common side effects — Headache; Nausea; Insomnia; Dizziness · Warnings — Seizure risk (dose-related); Neuropsychiatric events in smoking cessation; Hypertension · Interactions — CYP2B6 inducers; CYP2D6 substrates (e.g. metoprolol)
An item counts as shared only when both cards use the same wording. If a card does not list an item, that is not a statement that the effect never occurs.
